We have installed New TMG 2010 servers are not listing for port 443, we have 2 servers installed in same subnet. server can telnet at port 443 to self but can not do to each other.

Do we need to create any access rule in TMG to allow this ?. Please

yes, to allow access to specific ports on the TMG Server you must enable System policy rules or create custom firewall policy rules. In this case you must allow protocol HTTPS from INTERNAL or specific access to LOCALHOST for ALL USERS
