it is possible to upgrade windows server 2003 to 2008 & windows 2008 to 2012 R2 so after up gradation what ever application loaded on server  e.g SQL etc it  will work after OS is

Yes it is, just as long as in the 2008 to 2012 R2 example the 2008 installation is 64-bit, since there is no option for 32-bit installations with 2008 R2 and beyond, and no way to upgrade from 32-bit to 64-bit.

you mean if windows 2008 server 64 bit it is possible to upgrade windows 2008 R2/windows 2012 r2 i am right? second one is you mentioned "no way to upgrade from 32-bit to 64-bit" means what please explain with examples

Second one Keith meant to say that if you have a 32-bit operating system you cannot upgrade to a higher level 64-bit operating system, and you cannot reach to R2 unless you install as new.
