Hello,
Based on the research, Server Manager is servicing operations similar to what Windows Updates is using so the System Update Readiness Tool can help with this error sometimes.
To troubleshoot the issue,
1. Please first run the following command to scan integrity of all files on that problematic virtual machine
SFC /SCANNOW
2. Then, please run CHKDSK to fix the potential errors on the virtual machiner.
CHKDSK /F
3. Then please try running the System Update Readiness Tool to check the logs file to see if there is some error information.

Please note: the logs files (CheckSUR.txt and CheckSUR.persist) are under C:\Windows\Logs\CBS
If you see any error by using System Update Readiness Tool, please post back here.
4. Please take screenshot of the error message prompts.
Meanwhile, you can collect and send the log files and screenshoot to us via tfwst@microsoft.com for further research.
