Yes, BizTalk can be replaced  as a middleware between CRM Application and VSAM IBM mainframe . As a middleware tool BizTalk is being shipped with adapters which can connect to Both CRM and IBM mainframe.

You can Orchestrate your process by Using Orchestration and define business rules through BizTalk rule engine  .
