Please check the following: Do we have FBA (Form Based Authentication) enabled? Check the same under Exchange System manager>Name of the server>protocols>HTTP>Exchange Virtual Server's properties, then settings. Is there any certificate installed? Try to disable the FBA, and then try if you are still facing the HTTP 500 error? Note= After you disable FBA, please do an IISreset Check the authentications on the exchange v-dir, we should have basic and integrated only. Also check the apppool linked with the exchange v-dir, exchange application pool should be linked with the exchange v-dir Also try to browse the exchange v-dir from the IIS Manager and check if it works fine or you get the same error? And also try this article http://support.microsoft.com/kb/883380

Troubleshooting Outlook Web Access from an IIS Perspective http://support.microsoft.com/default.aspx?scid=kb;en-us;301428 If the issue persists, please check if there is any related errors in event log, if so, please paste here for our further discussion.
