I only have the 2 default send connectors configured when I configured EdgeSync (ADsite to internet and Internet to ADsite). Do i need to specify something on these connectors?

if user is created directly in the cloud you could have an issue with mailflow.
Can you try to send from a gmail/hotmail account to see if user receives it?

According to your description, I understand user host by on-premises cannot send message to Office 365.
If I misunderstand your concern, please do not hesitate to let me know.
As Off2work mentioned, please try to confirm whether all user host by on-premises can send message to external account.
How about the OWA?
Is there any firewall or defender software in your environment?
We can use Microsoft Remote Connectivity Analyze to double confirm external mail flow:

Meanwhile, run Message tracking log and post related error message for further troubleshooting.
