InfoPath forms are electronic forms that replaces the need to use paper forms. You can develop Vacation Request using these forms and submit them to your manager. To develop the workflow for the Vacation Request Action, you need to develop a workflow, this is where SharePoint Designer comes in, you can develop workflows by SharePoint Designer that your manager approves and then goes to the second manager and so on. You can consider SharePoint Designer 2010 as SharePoint Management Studio where you can do a lot of things using it, Applying master pages, managing SharePoint lists, External Content Types, and a lot of other things.

Visual Studio is where out of the box solutions and no code solutions made by SharePoint Designer are not enough, and you are in need to develop custom applications due to complex requirements, then you need to use visual studio. I suggest you know the out of the box features of SharePoint first, then know the capabilities of InfoPath 2010 and what it can do. Then start with SharePoint Designer and explore its features. After that you will what can be done with SharePoint and what can't be done using OOB features. Then you can manage using Visual Studio with SharePoint 2010.
