SCCM 2012 R2 with CU1

I enable multicast in DP, then if I need to click "allow this package to be transferred via multicast (winpe only)" of "Distribution Setting"?

Is multicast only support winpeor I need to click some checkbox for "allow this package to be transferred via multicast (NON-winpe )"

Correct. After enabling multicast on the DP, it also has to be enabled per OS image. This indeed can be done by selecting Allow this package to be transferred via multicast (WinPE only).

Also, multicast is indeed only possible when downloading an image file while being in WinPE.
