Ahmed, please review the following TechNet article and make sure that you've done all required steps:
Configure outgoing e-mail settings (Windows SharePoint Services)

few things to be verified -1> can u check how the SMTP is configured? 2> and also the creadential that is used in the configuration of SMTP? if there is a service account, then try to add the same to the sharepoint farm..3> also check if you can do a telnet on that to confirm if the SMTP actually works?Regards..

Great suggestions from ssm225in. Adding a couple more:- Try sending email from the command line on your SharePoint server (see example here: http://www.wikihow.com/Send-Email-Using-Telnet) - Enable SMTP logging and check the logs- Set SharePoint diagnostic logging for Email category to Verbose and check ULS logsAndre Galitsky, MCTS, Washington, DC --

Hi Ahmed,Make Sure that your Exchange accept relayed message from your Moss servers.what about configuring Relaying SMTP service, plz read the following:-
Configure the SMTP service
Click Start, point to All Programs, point to Administrative Tools, and then click Internet Information Services (IIS) Manager.
In IIS Manager, expand the server name that contains the SMTP server that you want to configure.
Right-click the SMTP virtual server that you want to configure, and then click Properties.
On the Access tab, under Access control, click Authentication.
In the Authentication dialog box, under Select acceptable authentication methods for this resource, verify that Anonymous access is selected.
Click OK.
On the Access tab, under Relay restrictions, click Relay.
To enable relaying from any server, under Select which computer may relay through this virtual server, select All except the list below.
To accept relaying from one or more specific servers, follow these steps:
Under Select which computer may relay through this virtual server, select Only the list below.
Click Add, and then add servers one at a time by IP address, or in groups by using a subnet or domain.
Click OK to close the Computer dialog box.

Hi Ahmed,
From your reply, you said that your e-mail server is the Exchange server 2007, which has been installed on a Windows server 2003.
Then when you configure outbound SMTP server, you need to specify the name of your Exchange server 2007. Next, the Exchange server must be configured to allow anonymous access and to allow e-mail messages to be relayed.
For more information about how to allow application servers to relay off Exchange server 2007, please refer to the following article:
Allowing application servers to relay off Exchange Server 2007

Hope this helps.
