I have a requirement to create a vanity url for a site collection. So is it possible to do in SharePoint 2010.

You can do this when creating a new site collection using hostNamed Site Collections.  But there is nothing that will provide this functionality for an existing site collection.  Here's an article on HostNamed site collections in SharePoint 2010.
