Claims-based site returns access denied when portalsuperuser and portalsuperreader accounts are set.

We have a 2010 Claims-based SharePoint system that currently works fine. Users can access sites and documents. We get errors in the logs about the portalsuperuseraccount and portalsuperreader accounts not being set. Whenever I set them, however, all users lose access to every site.

The user's have been setup to have permissions on the Web Application in Central Admin (Full Read for reader and Full Access for user). When setting the accounts up as the actual super accounts (I've used both stsadm and PowerShell) I do use the claims format of i:0#.w|domain\useraccount. As soon as I do so, I lose access to every site (and I'm a site collection owner) as do all my users. As soon as I unset the super accounts, access is restored.

Any suggestions would be appreciated.

Did you validate that the super user/reader are in Claims format on the User Policy as well? Did you run an iisreset after setting the super user/reader in the Web App properties?
